Fast and Efficient Separation of Mononuclear Cells (MNC)

Blood Separation Filter Tubes in 15 ml and 50 ml sizes are designed for the rapid and efficient separation of mononuclear cells (MNC) from whole blood by centrifugation. The tubes combine a specially designed filter with a density gradient, enabling simple, reproducible, and time-saving cell separation.

The method is based on ESQUIRE (Leuco-sep) technology and can be performed at standard room temperature without the need for temperature-controlled conditions. This makes the system ideal for clinical laboratories, hospitals, and research facilities where rapid sample processing is essential.

Reduce Processing Time from Up to 24 Hours to Just 30–40 Minutes

Using Blood Separation Filter Tubes, mononuclear cells can be isolated in as little as 30–40 minutes. Compared with conventional laboratory procedures, which may require up to 24 hours, this method significantly reduces turnaround time.

The rapid processing workflow enables laboratories to:

Materials and Construction

Blood Separation Filter Tubes are manufactured from high-quality materials specifically selected for laboratory applications.

Specifications

  • Available in 15 ml and 50 ml sizes.
  • Tube manufactured from non-cytotoxic polypropylene (PP).
  • Durable HDPE screw cap.
  • Integrated filter manufactured from PE-UHM (Ultra High Molecular Weight Polyethylene).
  • Filter pore size: 162–200 µm.
  • Sterilized using E-Beam irradiation.

Procedure

1. Prepare the Tube

Pipette Lymphoprep® into the Blood Separation Filter Tube.

  • 50 ml tube: 16 ml
  • 15 ml tube: 3 ml

2. Initial Centrifugation

Centrifuge at **1000 × g** for **30 seconds** at room temperature.

3. Add the Blood Sample

Fill the tube with freshly collected anticoagulated blood.

  • 50 ml tube: 15–30 ml
  • 15 ml tube: 3–8 ml

The blood may be used undiluted or diluted directly in the tube using a balanced salt solution.

4. Cell Separation

Centrifuge at either:

  • 1000 × g for 10 minutes, or
  • 800 × g for 20 minutes.

Following centrifugation, the sample separates into the following layers:

1. Plasma
2. Mononuclear cells (MNC)
3. Lymphoprep®
4. Filter
5. Lymphoprep®
6. Erythrocytes

5. Collect the Cells

Carefully collect the MNC layer using a Pasteur pipette or gently decant it into a clean centrifuge tube.

6. Wash the Cells

Add 10 ml PBS, gently resuspend the cells, and centrifuge at **250 × g for 10 minutes** at room temperature.

7. Repeat the Washing Step

Repeat the washing procedure twice and finally resuspend the cell pellet in 5 ml PBS.

8. Ready for Storage

The lymphocytes are now ready for downstream analysis or cryopreservation in suitable cryogenic tubes.
